He was born as Thomas Luke Macfarlane Jr. to father Thomas Macfarlane, the Director of Student Health Services at the University of Western Ontario, and mother Penny Macfarlane, a mental health nurse at a London hospital.
He is not the only child of his parents and has two siblings- older sister Rebecca Macfarlane and fraternal twin sister Ruth Macfarlane.

The aspiring actor started his career in 2003. He was one of the four leads in a theater production of Juvenilia in 2003 followed by the lead role in the 2004 theatre production of Where Do We Live.
And in 2004 Luke Macfarlane landed a minor role in the film Kinsey. This was followed by a minor role in the 2004 TV miniseries Tanner on Tanner and in the 2005 TV series Over There as the main character Frank "Dim" Dumphy.
Finally, in 2006 he was cast in the hit drama series Brothers & Sisters as Scott "Scotty" Wandell. He portrayed the character for a total of 81 episodes.

Since his breakthrough role, Luke Macfarlane has had appearances in films Trapped Ashes (2006), and Rock Paper Dead (2017). He has also starred in the TV series Satisfaction (2013), The Night Shift (2014-2017), Killjoys (2015-2019), and Mercy Street (2016-2017).
