His first roles were in the films Heaven's Soldiers (2005), The Moonlight of Seoul (2008), Take Off (2009), Midnight FM (2010), The Unjust (2010), etc. He also has appearances in the episode(s) of the TV series H.I.T (2007), Robber (2008), Swallow the Sun (2009), Dr. Champ (2010), to name a few.

Other films in which Ma Dong-seok has starred include Perfect Game (2011), Dancing Queen (2012), The Neighbor (2012), Rockin' on Heaven's Door (2013), Marriage Blue (2013), The Royal Tailor (2014), Deep Trap (2015). Finally in 2016, his role of Sang-hwa in the film Train to Busan is considered his breakthrough role. His following film roles were in the films The Outlaws (2017), Along with the Gods: The Last 49 Days (2018), Ashfall (2019), and in the Marvel Comic film Eternals (2021).
