Richardson’s debut role was in the 2001 film Flossin' as Malik followed by minor appearances in 7th Heaven and The Bernie Mac Show, both in 2006. He then went on to work in the TV series Unfabulous (2007), ER (2008), The Starter Wife (2008), web series My Alibi (2008-2009), True Blood (2012-2013), The Newsroom (2013), among others.
Finally, in 2014 Marque Richardson was cast as Reggie Green in the comedy-drama film Dear White People (2014). It gained him some recognition.

He partnered in 2018 to form The Jordan Edwards Memorial Scholarship Fund. They award scholarships to honor the legacy of Jordan Edwards, a child who was killed by a police officer in Dallas, Texas.
Back in 2005, he traveled to Khao Lak, Thailand to provide relief after a tsunami in December 2004.
