Middleton landed her debut role in the TV series Bones in a 2008 episode. This was followed by the role of Justine Fielding in the 2009 comedy horror film Tormented and her breakthrough.

Over the years Tuppence Middleton has starred in over a dozen films. Some are namely Chatroom (2010), The Love Punch (2013), The Imitation Game (2014), Jupiter Ascending (2015), The Current War (2014), Downton Abbey (2019), Possessor (2020), and upcoming film Mank as Sara Mankiewicz.
Moreover, Middleton has had guest appearances in the TV series Sinbad (2012), Friday Night Dinner (2011), Black Mirror (2013), and in the miniseries War & Peace (2016). Also, Tuppence Middleton has been cast in the main role of Riley Blue Gunnarsdóttir in Sense8 (2015-2018) and as Amelia Havisham in the short-lived drama TV series Dickensian (2015).
