Luke began his career as an actor in 1999 in two episodes of The King of Queens (1999-2000). He was working at a store, folding clothes then. Finally, in 2002, he landed his breakthrough role as Antwone "Fish" Fisher in the film Antwone Fisher.

This was followed by roles in the hit films Glory Road (2006), Catch a Fire (2006), Miracle at St. Anna (2008), Madea Goes to Jail (2009), Captain America: The First Avenger (2011), Baggage Claim (2013), Supremacy (2014), Extraction (2020), and Rare Objects (2023).
