His mother is Christina Daddario, a lawyer and his father is Richard Daddario, a prosecutor and former head of NYPD Counterterrorism. His parents raised him in New York with his two siblings- younger sister Catharine Daddario and older sister Alexandra Daddario who is also a successful actress.

This was followed by roles in the films Breathe In (2013) as Aaron, 36 Saints (2013) as Sebastian, Delivery Man (2013) as Channing, When the Game Stands Tall (2014) as Danny Ladouceur, Naomi and Ely's No Kiss List (2015) as Gabriel.

Matthew Daddario’s breakthrough came in 2016 as he was cast as the main character Alec Lightwood in the supernatural drama series Shadowhunters. He portrayed the character until 2019.
