Murin began her career as an actress in 2002 with the theater production of Crazy for You as Polly Baker. Patti Murin went on to star in a theater production of Parade (2003), Annie Get Your Gun (2004), Mirette (2005), and Beauty and the Beast (2006), among others.

On Broadway Patti Murin has portrayed characters in Xanadu (2007), Lysistrata Jones (2011), Frozen (2018) and off-Broadway Lysistrata Jones (2011), Fly By Night (2014), etc.
