Born as Enrique Ricardo Murciano Jr. to mother Christina Murciano and father Enrique Murciano Sr., he has two older sisters. One of his siblings Caro Murciano is married to Mayor Philip Levine, former Mayor of Miami Beach, and has three children - Mica, Henry, and Selma.

Murciano began his professional career in 1997, his debut role being in the film Speed 2: Cruise Control (1997) as Alejandro. This was followed by roles in the films Traffic (2000), Black Hawk Down (2001), guest appearances in an episode of the TV series Suddenly Susan (1999), The Pretender (1999), Star Trek: Enterprise (2002), and the main role of Francisco Torres in Spyder Games (2002).

Finally, in 2002, Enrique Murciano was cast as Danny Taylor, the main role, in Without a Trace (2002-2009). He portrayed the character for a total of 159 episodes and is considered his breakthrough.

Over the years he has starred in films Miss Congeniality 2: Armed and Fabulous (2005), Dawn of the Planet of the Apes (2014), Rough Night (2017), The Half of It (2020), and in the TV film The Saint (2017). 

Moreover, Enrique Murciano has had recurring appearances in the TV series NCIS (2011-2012), Power (2014-2016), The Blacklist (2017), and the main role of Marco Diaz in Bloodline (2015-2017), etc.
